My most significant achievement was to establish conjugation with Small Ubiquitin-like Modifier (SUMO) as an important regulatory mechanism in eukaryotes. My lab (and the Blobel lab) were first to demonstrate that the enzyme involved in SUMO conjugation was Ubc9. We demonstrated that the acceptor lysine for SUMO modification in IkBa was also the acceptor
for ubiquitylation. This established the paradigm of SUMO and ubiquitin modifying the same lysine residue, allowing proteins to switch between different modes of activity and regulating many biological processes.
